I am interested in PC games that can be played with multiple players on a single machine, but unfortunately it looks like there aren't a lot of those available.
There are a few games that I can think of already, but I want more:
Lego Star Wars/Indiana Jones/ Batman
Marvel Ultimate Alliance
Worms (and most other turn based games, I guess)
Yeah, this is extremely old, but you might be able to find a copy of Robot Rascals somewhere to play with DosBOX. It's the only computer game I've ever played that has a deck of cards and 4 players required.
Just thought I'd throw out an oldie that I'm sure would be overlooked.
I think all the LEGO *** (Star Wars, Batman, Indiana Jones) do it.
I'm already aware of the Lego games, which is why I mention them in the OP. I'll look into Serious Sam, though. Is that available on any digital distribution services?
I looked at Capcom's website for Bionic Commando Rearmed, which says it has up to 4 players and no online, so that must mean it's on one machine. Now that I have a computer that can run it, I'll get it anyway as I hear it's awesome.
You can also play games such as "hotseat" Battlefield 2 where you switch places upon respawning.
Note: There isn't a hotseat mode, I'm implying it be a homemade rule.

I have never used Direct2Drive and I can't actually GO to that link right now because my work proxy is pretty good at catching game links. I'm going to check that out at home though because I've been meaning to get my hands on a legal copy of this for some time (lost my pirated Second Encounter long ago). Additionally, you can just grab it from Amazon under $20 here, a huge steal I think. I bet this could sell well re-released on a current-gen console...
Edit: oh, right. Civilization is another good one. FreeCiv, if you're cheap.
Edix2: OH! Sonic 3 & Knuckles Collection! My sister and I spent hours playing through as Sonic and Tails. Sonic R is also fun, although it's highly imbalanced and has not aged well at all.
Spent a lot of time playing this with my brother years ago. Bonus points for level editing/scripting.

If you have or can acquire any of the Gold Box D&D games and feel like messing around with DOSbox, then they provide a fine multiplayer-ish experience. I used to sit around with a friend and we'd split the party between ourselves, making those characters our own.
I imagine that this could work well with games like Jagged Alliance as well. Other players input during combat made for interesting situations and some good hilarity when things go wrong.
The Ur-Quan Masters (i.e., Star Control 2) has a 2 player mode that was entertaining for a little while in 1992 or thereabouts.
Co-optimus lists Bionic Commando: Rearmed as having offline split screen.
EDIT: It occurs to me that M.U.L.E. was basically the best offline PC game ever made. Wikipedia says there was a clone called Subtrade that came out in 1993. And there's another clone called Traders that might also be worth checking out.
I also tried looking for Archon clones, but I don't see anything more recent than Dark Legions and Archon Ultra, neither of which I think has aged well.

Oh, also, X-Men Legends 2 is multiplayer.
You can also play Left 4 Dead splitscreen, that's just a tweak to enable it.
I actually have that. I guess I should install it.
Heroes is good, but how long it takes to play is inconvenient.

Besides turn bases games, that's really all I know of.
EDIT: Holy shit how did I miss that? Beat'd.
Still awesome though.
Yeah, they do. At least First and Second Encounter do, I'm not sure about Serious Sam 2.

Yeah, if you have a spare gamepad of sorts around then Serious Sam co-op is all sorts of good times.
(It did not work well so yeah a gamepad makes things cool)
I did not know this. Investigate, I shall.

Just make sure that you have a gamepad (preferably PlayStation 2) to use while playing it.
And I also heard that Rayman Origins is coming out on PC, maybe that might have local/online multiplayer, unlike PS Vita. >.>
I second Rayman Origins, as it's excellent.
Also, this thread needs more love for Jamestown: Legend of the Lost Colony

You Don't Know Jack is a freaking great party (trivia) game.
