you might just want to ask/read up over in Artists Corner.
I'm pretty sure that a bunch of folks over there rock them. They had a bunch of stuff in the tutorial thread about them, but it looks like that was a few itertaions back cause the current on is down to 10 pages.
They are a bunch of hermits and tend to not veture out of thier little hole, but I doubt they would mind you asking over there and you're likely to get more of a response.
Majority of the time he sketches something and then scans it, inks it with his tablet and then proceedes to colour it and add finishing touches.
Now what he was also talking about is how with the CTS comic he will chose to do soemthing differently. I believe he made note of the fact that he scanned his hand inked comics and then coloured them to give a more classic style.
But yesh most of the time Gabe sketches, scans, inks and colours in phototshop jusing a Wacom tablet. I forget if it's him or Hawk(From applegeeks) with the Cintiq tablet.

I think Gabe did the comic in Photoshop, but was playing with with Sketchbook and talking about how cool it was when he was on the road. Back during 'Gabeart mark I' I beleive. I could be totally off my mark though.

I thought they sketch it on paper, scan it, and color it using a Wacom tablet and Adobe Photoshop.
I could be wrong, but Gabe posted a videoblog of bringing one of his strips to fruition a few weeks ago and I believe that was his process
NOTE: I'm entirely unsure if that is the process they use all the time.

Now, the artist has this thing called "talent". You can't buy that. The only way I know to obtain this item is through lots of practice.
Tip: Draw from life, not cartoons.
