MEAT THE CAST:
Coach: A high school football coach with a bad knee, and a craving for chocolate.
Rochelle: A news reporter from a local television station, as well as a snazzy dresser and fan of Depeche Mode.
Ellis: A mechanic who has probably never heard of cricket.
Nick: A gambling conman, and some kind of cliche movie quote machine.
All the survivors play exactly the same, so don't get mad because you didn't get to be Ellis. Even though Ellis is the best and it's quite understandable you would be upset about that.
There are brand new infected this game, as well as some modified versions of the originals. Not counting the snazzy new models, of course. All of which are playable in versus, except the witch.
The Boomette: There's now a female Boomer, the only female version of the original infected.
The Wandering Witch: Witches now wander in broad daylight, and are much harder to avoid.
The Spitter: A zombiefied American Apparel model, she spits out a lethal green acid. She also leaves a pool of it around her corpse when she dies, and damage increases the longer you stand in it. The acid does not harm other infected.
The Jockey: A monkey-like infected that can leap onto survivors and steer them away from the group, even into fire, acid, and witches.
The Charger: A large, almost half-tank infected, who can steamroll survivors and pin them to the ground. He'll continue pummel the survivor in his grasp until stopped. He also does more damage than the other special infected that "pin" their enemies.
There are also special common infected, like zombies wearing hazmat suits or riot gear. There are also a couple of others, but we'll let you be surprised.

There are also new game modes in addition to Versus and Campaign, Scavange Mode and Realism Mode. (copied from the L4Dwiki)
In Scavange Mode, the survivors have 2 minutes to find 16 gas cans hidden around a map. For each gas can they retrieve they gain an extra 20 seconds. The goal is to fill a generator with the gas found amongst the 16 canisters while fighting against player-controlled Special Infected much like in a versus game.
Realism Mode can be played on any difficulty setting. This mode removes the auras usually shown around players through walls. Also, the auras around items are only visible within a few feet, making the game require more communication and teamwork. Realism mode also makes bodyshots less effective at dispatching Infected, while making headshots more effective, making headshots the only efficient way to kill infected. Survivors will die outright instead of being incapacitated when they run out of health, and rescue closets are removed, making the defibrillator the only way to revive dead teammates. Additionally, attacks by startled Witches will instantly kill Survivors on any difficulty setting. Between the dependence on headshots and the number of Witches around, melee weapons are even more useful than usual.
