I've just gone and got myself a Playstation 3 and thought you fine people here could help me along the line of unmissable games.
I've grabbed Heavy Rain, Little Big Planet and Valkyria Chronicles (and plan on getting Demon Souls and 3D Dot Game Heroes). I already own a 360, therefore I'm really only interested in PS3 exclusives or muti-platform games that are generally regarded as better on the PS3.
Pixeljunk Racers, Monsters, Eden and Shooter on PSN. I think the first three have demos. Racers is solid but can get pretty ridiculous (play it and you'll know what I mean by that). The last three are phenomenal.

I enjoyed Heavenly Sword. It's a bit on the short side, but it's a Platinum title (cheap) and a quality product for the most part.
Fuck the shooter sections, though. Fuck 'em with a battle axe.
edit: Free tip: Turn off the shit awful motion controls.

Pixeljunk Monsters is especially worth it if you have a friend who enjoys tower defence too. The coop is really good.

Resistance Fall of Man. It's cheap now, and it was a really good console FPS. It's also about as long as 3 current gen games stuck together. It just keeps going and going, great value. Decide after that one if you want more, but Resistance 2 is a lot different.
The Ratchet and Clank games. Seriously, just do it.
God of War 1, 2 and 3. Play 1 and 2 first, otherwise they'll be extremely underwhelming after 3.
I liked FF XIII. If you like it you should get that. You would already know if you're into those types of games, but it's probably going to be the first FF game I completed since 4.
Wow that's some great suggestions there. I'm definitely tempted by a good portion of them.

Everyday Shooter! It is a delicious beautiful take on something like Geometry Wars. Every level is a song, and has completely unique game mechanics, and it looks and sounds gorgeous.

I never played the first three MGS games so I found myself completely lost as to the plot, but I was still able to enjoy the gameplay immensely. From what I saw though, I wasn't missing much of a plot.
With MGS4, would I need to have played the others to follow the game? (I've played the MSG Remake on the GC but the others have passed me by).
The MGS games are some of the most continuity heavy games around, but at the same time...the plots are really pretty bloated and dumb overall anyways. Most of the enjoyment comes from the presentation of the plot and fun stealth gameplay.
I'd say go for it anyways: you'll be confused, but that seems to be a central goal of the dev team for those who have played them all too.

A bit of research on some key characters that carry over game to game like Naomi, Meryl, Otacon and Liquid and he should be right to go. That's all I did, and I wasn't lost at all. There's a lot of stuff that references the previous games, but overall the plot is pretty easy to follow by itself. Smoe of the big reveals won't make an impact though.
Couldn't give enough love to the Ratchet and Clank games. They are awesome. Both Tools of Destruction and Crack in Time have demos on PSN if you're unsure.

Infamous is a fantastic game that just seems to have dropped off everyone's critical radar, despite good reviews at the time, which is a shame. I loved it.
Demon's souls is a vicious bitch that tears out your heart time and time again but its just so unlike anything else this generation.

Edit: I picked up a used copy of MGS4 on the extremely cheap. After a few hours I decided it was not for me (never really played any games in the series) and it was later traded in as part of a downpayment on Uncharted 2. Best trade I ever made :P

Well, I think its a pretty bad game. Slick? It has a horribly clunky combat system (three different fighting stances? An auto-block? some sort of bizarre colour-coding system for enemy attacks?), the puzzles are a joke (a box of hats! Woop!) The art direction is terrible (medieval europe mixed with half-baked oriental fantasy where everyone speaks in comedy cockney or scottish accents. Uh huh.) and they clearly blew all the money they should've spent developing the game on those terrible cutscenes. And the story takes itself too seriously.
Plus its about six hours long, cutscenes included.
Yes I bought it full price. If you must play it buy it cheap. It does look nice i'll grant, though the framerate gets a bit choppy at times.
[edit] Christ, I forgot Wipeout HD. Totally essential with the fury pack.

On this style of game though, the OP should definitely get Darksiders. It led on PS3 and is better on it.
#1 Favorite: inFamous. I'm not kidding. There are games of higher overall quality, to be sure, but no game this gen has provided gameplay of such uniformly high pleasure than inFamous.

That's Uncharted 2. I'm not kidding. During my first playthrough, I was so giddily thrilled with the experience I found myself laughing out loud. Not at a joke, or a clever reference - simply because my Cup of Happy was bubbling over, and had to be released through exultation.
Siren: Blood Curse.
Siren is the single most under-appreciated game on the PS3. Far too few have played it, but even worse far too few even know it exists.
Wonderful art direction and atmosphere, absolutely incredible music, classic adventure gaming tropes, the best damn variation on the shambling dead I have ever seen and - here's the kicker - built from the ground up as a stealth game.
Consider that. The already stress-fueled survival horror genre combined with the tense, nervous gameplay of a stealth title - but Siren's genius isn't limited to its loving combination of mechanics and styles. It is as smart as Silent Hill 2, and twice as creepy. Oh yeah. I went there.
Also recommended: Ratchet & Clank, Resistance, Heavenly Sword (pretty much just for the presentation - still worth it, if you can find it cheap), God of War III, Heavy Rain, Killzone 2.
Edit: Oh, and Mirror's Edge, Burnout Paradise and Darksiders all lead development on the PS3, so it's the way to go, there. I've also heard if you intend to pick up FFXIII to get it on PS3, but I'm not about to endorse or condemn it either way.

inFamous was kind of fun for a bit. Everyone spanks over Uncharted but I hated it.
Darksiders is fun and better on the PS3.
I liked Folklore a lot. Eternal Sonata is better on the PS3 but jury's out as to whether it's worth playing at all.
Apparently Burnout Paradise is much better on the PS3.
And yeah, avoid FFXIII at all costs. It looks amazing, but is one of the, if not the, worst game I've played this entire console generation.
Edit: And whatever you do, don't forget to set aside money for the Last Guardian, the third Team ICO game, sometime before it comes out.

Get FFXIII, it's one of the best RPGs of all time.
