I am a gamer girl. And by that I mean I have a vagina and also play games. I don't play games about cooking, making jewelry, getting boyfriends, or taking care of animals/babies. I play D&D, Fallout, Magic the Gathering, Mass effect, halo, a variety of board games (such as Dominion and Settlers of Catan, not Candyland), among many other games that don't come in pink packaging. I don't flirt/sleep with men in return for games. Being a girl doesn't make me incapable of being a gamer, nor liking certain games, nor having any form of basic intelligence. I am a human being just like the male of my species and feel no need to constantly gossip with others or cry when I don't have someone constantly asserting their physical attraction to me.
My father and step-mother both work in the games industry. I have been immersed in that culture my whole life. I don't doubt the creators of Penny Arcade, at least on some basic level, acknowledge the difficulty of being a girl in gamer culture as well as the hardships that come with being a woman working in the games industry. Then, why would Penny Arcade make such a tasteless video that perpetuates stereotypes that annoy the hell out of girls like me on a daily basis? Penny Arcade, I am disappoint.
I don't think a thread where your OP is basically you scolding will end up well. Even though I'd agree gaming as an industry and culture has a ways to go towards female consideration.

Presently, there are two choices--either accept the assumption and the benefits thereof (lack of female gender stereotypes), or announce gender and accept the consequences (in this case, negative stereotypes).
I think the ideal is that it shouldn't be assumed that a gamer is male or female--some people already do this, but I think we can accept that generally the gamer is assumed to be male. Hiding gender while playing isn't likely to change the assumption. If you're going to make it your cause to change the assumption and negative stereotypes that go with being a female gamer, then that is entirely your choice, but be prepared to face the backlash of the image of groups like Fragdolls and GameCrush and the negative stereotypes until those things change.

Of course by announcing one's existence out of the assumed characteristics opens one up to negative perceptions, but it also presents the opportunity to challenge those perceptions.
Besides, it's not like most (MOST) people go around with usernames like GAY GAMER HERE or HERE BE BLACK FOLK, it's more like when someone says something, like, assuming you are a guy whose girlfriend or wife will aggro you, you say, "nah, actually I'm a woman, my husband's doing the dishes."

I didn't find it funny. It was pretty tasteless, especially if cross dressing is trying to get laughs. But I think the larger statement is attempting to make fun of the gender difference between male and female gamers (i.e. girls being hyperfem and guys being ubermacho). I honestly don't think it did a good job of it, so the OP has some merit in not liking the result, but it wasn't malicious.
Did anyone actually watch the ending, since it hasn't been mentioned once? If you didn't watch the ending, then watch the ending, because it makes more sense that way.

Two out of three of those are visibly obvious from a distance, though, and one is aurally obvious the second I get on TS or Vent. The remaining item becomes obvious the second I say, "brb my husband/wife just brought me a drink."

I suppose there's an argument that stereotype lampooning can still be harmful. Imagine if Robert Downey Jr's character in Tropic Thunder had acted like Stepin Fetchit. There'd probably be people angry if they saw Al Jolson in blackface today, even though he was anything but racist.
