You and 39 other players are thrown together in a town in the middle of the wasteland. Every night at midnight (in my games it has been 4pm CST) a horde of zombies attack the town. For every point of defense you have, you block 1 zombie. You have to work with the other players to build the town up, including improved defenses or other upgrades that help you in various ways. Each night the attacks usually grow stronger and stronger (not always), so you constantly have to be improving things. This requires quite a bit of coordination between the 40 inhabitants of the town.
Each day (a real 24 hour day) you get 6 action points to work on projects in town or wander around the wasteland searching for supplies. Doesn't sound like a lot, but searching the wasteland is free and moving around the town is free. What you actually spend points on are walking around the wasteland (1 Action Point per square), and labor projects in town (building a pump for the well costs 25 AP, you basically contribute however many you want).
You can replenish your AP in a number of ways, the easiest way is by drinking water, which gives you back 6 AP. There is a town well and as a citizen you get 1 water ration per day. If you don't drink at all you will get thirsty and then dehydrated and then die, so you have to drink water regularly. The town has a limited supply so you either need to upgrade your well to get more water or find water in the desert.
You can also eat food to replenish AP, but this recovers a variable amount. Some foods actually give you more than 6 AP (steak gives you 7). There are also questionable things you can eat which can have negative side effects if you aren't careful.
You can also drink alcohol to get your action points back, or take drugs, but these have serious side effects, such as the chance to get addicted, or hungover.
So you can walk 6 spaces out into the desert, search for several hours, then drink water and walk back. You could also put 6 action points into a project in town, drink water, then put 6 more, eat food then put 6 more for a total of 18 points.
The really great part of this game is the interactions between the citizens. Anyone taking more than their share of supplies or otherwise acting recklessly can be banished from the community by a vote. They still get to stay within the walls but they are denied access to the well and the bank (they can put things in but can't take things out). Every area of the town has a message board where you can talk to each other, and communication is vital as you need to plan out your building strategies based on what materials you get from searching the wasteland. It's also fun when someone notices a player hoarding supplies and they gang up on them, banishing them quickly. You can also lock people outside of the gates, where they will likely die (supposedly there is a way to survive outside after midnight but I don't know how yet or they haven't implemented it yet).
If you die, you're gone. Your town goes on without you. You can immediately join a new town that is just starting up.
When you feel that a random town is a bit too much for you then you can join up in a coalition with up to 5 players that will join a town together.
There is also a premium "Hero"-mode that give you a variety of in-game advantages at a cost but you can also get a couple of days of "Hero"-mode by being the last man standing in your town. If you wish to sign up then you might as well use a

I do the free-2-play option. If you form a coalition then your members auto join with you if they're not in a current game. So coalition members play together regardless of p2p or f2p.

It could do with a price cut down to facebook-game prices (I've never played farmville but that's a couple of bucks a month right?)

Towns can now execute thieves so you better watch out
