● Greater variety of mission types
● More buildings with fully accessible interiors
● Better check-pointing system (BoGT fixed this, mostly)
● A safehouse with a helipad
● A seaplane/Heli that can land on water, like in VC
● Heli with a winch/magnet, just to pick up cars and mess with peds
● Even more ped behaviours, people just going about their daily lives, more police/ped interaction, other peds comitting crimes etc, random events like in RDR.

GTAIV was a huge game, with a truly living city. Yet it was a huge downgrade in terms of "stuff to do", replacing exciting things like jetpacks and Area 51-raids with bowling and your cousin. No more God damned dating, playdates and people calling me for darts. I mean, I guess I don't care if it's there, but there's sometimes bonuses to reaching certain "like-evels" and I'm a terrible completionist. I gave up 100%-ing GTAIV just because I hated the playdates so much.
I'm liking that GTAV looks more vibrant, which is great, because that's a large part of what I want from GTA. Hopefully vibrancy means some of that trade mark over-the-top, cartoon-ish humour makes its way back into the series. Extremely doubtful, but I like my GTA less dour than IV was.

AI crew. A good one you can customize, not the 2 dumbasses that Niko called up with a pistol and an Uzi, or some weak motorcycle gangsters. Something like GF:TG did. Territory metagame like San Andreas.
i really want to know if it's just california, or if they are gonna do something like san andreas with lots of different areas.

Going back to the hood with a harrier jump-jet and just wrecking everyone's shit was the best thing. The only thing that could have made it better would be destructible buildings.
I want to see something less full of pointless grit and angst than IV, but not quite as over the top crazy as SR3.
GTA 4 has actually aged really badly. The city is incredibly well realized but that's pretty much the only nice thing I can say about it. The cover shooter gameplay is really, really bad and almost every single mission is totally linear. About the only missions where you have any freedom in your approach are the phone box assassination missions.
I respect rockstar for having their own vision with what they want to do with GTA but it's pretty blatantly the wrong vision.
Also the storyline is kinda bad. I really hate the unearned pathos when you play a psycho (admittedly a likeable psycho) and he's all like "I wanted to get out, live a normal life" and then some drunk is like "kill this guy, he owes me $20" and Niko is like "sure okay" even though he already has a nice suit and a penthouse apartment.
Less of a disconnect between the storyline and the stuff you can/must do in the game.
I actually kind of like the idea of hanging out with other NPCs, but I'd much prefer it if I had to call them to set it up instead of the other way around. That way, at least, I have some control over when I go bowling and who I go bowling with.
Ballad of Gay Tony but longer, I really enjoyed those missions and the story. Need to get Brucie and Yusuf together. Never ended up finishing Lost & the Damned though, something about the motorbikes didn't do it for me

Following Saints Row 3, I hope they avoid the temptation to bring it back to wacky fun times. Don't think there's really room on the market for 2 dick around sandboxes, I'd much rather they go full tilt in the other direction, play it all straight - no jetpacks or shark guns or anything of the like.
Honestly, after playing Sleeping Dogs so much, pretty tempted to just point to it and say "that but in San Andreas"

I want to have more stuff to do in the sandbox gameplay that isn't missions.
GTA has always suffered from "you can drive anywhere and do anything but outside of random acts of violence, there's nothing to do except missions"
They should take a leaf from Just Cause 2 and have random things you can do.
Just don't make the story be a complete contradiction of the actions they have you perform. They got it right with Episodes from Liberty City, so I'm confident they'll get it right again. In fact, the most interesting rumour I've heard is that GTA V will feature multiple protagonists. I think that's excellent since one of the reasons I vastly prefer the episodes to the base game is that they have a tighter and more focused story.

Mainly I want it to have a lot of desert/countryside (even if it excludes San Fierro and Las Venturas) and some good radio stations. More stuff to find while exploring (whether they be side missions or just weird sights) would also be cool.

I kind of want the opposite. Saints Row 3 was going in the right direction in terms of tone, but it lacked GTA's follow-through---the city felt dead, uninhabited, compared to a GTA city. I don't think GTA needs dildo weapons, but some more wacky fun times would be welcome. The gameplay is over the top by its very nature. It doesn't work to fight or ignore that.
I know they've already shown some shots so it isn't happening but I want a whole game in the Las Venturas (Las Vegas) section from San Andreas but set back when the town was run by old school mobs. I want to start as a lowly thug in a gang and work up to owning and running a casino.
Characters and vehicles that don't move like they're made of stone would be nice. Everything just felt so weighty to the point of being horribly ponderous that it just kills the game for me (and LA Noire too as I unfortunetily discovered during a recent sale). RDR was so much better in this regard and considering its success I'm hoping they're integrating some of its design philosophies into GTA V.
I want more light/fun moments, but no unbelievable wackiness. No Area 51, no jetpack, no "Street hood can fly a military jet" stuff.
That said, there definitely need to be more vehicle types, across the board. I really want a jet-ski.
I also want to see more easter eggs and rewards for exploration. IV was incredibly disappointing in that regard.
And, of course, what others have said: more building interiors, better NPC AI, more things to do (I'd love it if we could play tennis), better safehouses (another thing IV failed at - only two nice ones, and one was only available if you killed Playboy X), etc. I'd love to own property again, too.
Take the random encounters from RDR, ramp them up to eleven, add a million of them, ditch the hamfisted narrative and make those random encounters in the city the game.
That and muhfuggin building interiors.

A cop happens to be walking by and he engages the thugs, calling for backup. More cops arrive and soon they're swarming into the house where a massive firefight is taking place.

UN in RDR was so fantastic. Zombies in a modern GTA open world setting would be pants creamingly amazing.
