I was surprised by the "new galaxy" thing, but I understand it. The endstate of the old galaxy is
a disaster, both diagetically and otherwise. There are huge, setting-defining questions that the writers never attempted to address, like "are certain entire civilizations dead?" and "is everyone just gonna starve now?" Trying to set a game in that particular gordian knot would be a nightmare.
I wouldn't get too suspicious at the old west stuff, I think that was a trailer choice more than anything else.
My favorite bit was that Vigil-sounding music at the end. Man I am a sucker for the Vigil theme.

I can back the accuracy of this statement. According to recent studies, somewhere between 75 and 80% of Mass Effect players are human, allowing the standard 5% margin for two sectoids in a trenchcoat.

My understanding is that the crew of this colony ship has actually been on this mission for several generations, due to the long time it takes to cross the vast distance between The Milky Way and Andromeda. They're probably still using the same hand-me-down equipment their forefathers brought with them when they left.

can you elaborate on what you mean? Because cheesy romance subplots and hero stories kind of define Bioware games and I'm not sure you want to fuck with that kind of formula success.
Biorware's stories almost universally go like this:
Introductory/tutorial level/establishing that you are the mysterious stranger/introducing twirly mustache
Hub zone I have to slog through for a few hours before I'm allowed any actual freedom of exploration of the greater universe/world
3 locations I need to explore (nominally connecting to you being the mysterious stranger/ twirly mustache is super evil)
zomg twist
love scene
final battle with twirly mustache
make a final choice
credits
I really want them to break away from this formula, especially since the "hard choices" are usually whether you want to be a douchebag or not.
Hell, it probably goes a long way towards why I like DA2 so much more then the first game.

I'm curious as to what the impetus (in-game) is to colonize the Andromeda galaxy. Is it a Plan B for the Reapers, Humanity (and Krogan?) tired of the Council's bull, or more of a general space exploration/Star Trek thing. It'll be interesting to find out what they have planned.

I'm really hoping they crank the wild west/frontier feel to this up a notch. I love that sort of aesthetic and that, coupled with the whole 'Exploring, prospecting and settling a new Galaxy' thing seems like a natural fit.
Hell, if he's human, give the main character a Texas drawl. Gimme armor and the like with bandannas around the neck, and satchels.
