Concerning Castlevania:
Dawn of Sorrow if you like exploration and lots of zany abilities.
Order of Ecclesia if you like relatively straightforward games that are hard enough to melt your face off.
Portrait of Ruin is middle ground, not as good as either of the other two, though still pretty good. With an awesome last boss.
Portrait of Ruin also has the most 'meat' to it - half the bosses from the first game, Dawn of Sorrow, are hidden in the second game and boss fights are, of course, the best part of the Castlevania series. :P
Slashsignal's recommendation page (posted earlier by lowlylowlycook) is great. I just recently picked up both of the Trauma Center games, and they're great. You can get the 2nd one brand new for like $14 from Amazon, but you'll have to get the first one used (Gamestop has them).
New Super Mario Bros. is good if you love your Mario games.

I nominate Disgaea, Mario Kart, Pheonix Wright 1, and Brain Age/training for it's awesome sudoku.
One of the best looking and most fun games on DS. It's an absolute shame this game has not gotten a sequel.

Not to turn this into a Castlevania thread, but all of them are good, all of them have weak points and strong points. I'll hide it here in a spoiler and get on to other recommendations.
DoS' weakpoint is that you see little change in it from AoS, aside from multiple souls actually doing something to most abilities. It's music is ho-hum. It's super-duper anime in how characters act.
PoR has a really strong soundtrack and has more in relation to canon in the series than the other two. It's weakpoint is not-as-good level design.
OoE is probably the strongest of the three DS titles. Great soundtrack, there isn't a clutter of items at all (maybe the accessories, but barely). If there's a weakpoint, it would have to be Shanoa's personality.
If anyone tells you to pass up on any of the games, they're stupid. I try to refrain from saying such things about opinions but come on now.
Edit - Oh, another strong point about PoR and OoE (but moreso PoR) - the english voices and such for enemies are actually varied quite well and takes many many steps away from the bullshit anime "HIYA" and "HA!" kind of crap that you'd hear in most games prior in the series. But if you want you can switch to the Japanese voice track. I consider this a strongpoint because it shows effort in enemy design. And I'm tired of heavy anime themes.
If you like oldschool RPG leveling systems and brutality, get Etrian Odyssey (I would say get part one for perspective, and then get part 2 for even more awesomeness). Final Fantasy 3 isn't too bad either.
You can't go wrong with the Ace Attorney series (and for the love of God, if you do get into them, start with the first, because the continuity matters).
I'd recommend Mega Man ZX if you want a taste of Mega Man series, but this game is kinda goofy. It's hard to navigate your way around to levels, since it's a large map to explore, and the weakpoint thing on bosses has some people frustrated because it affects power meters for your other abilities. I haven't played the sequel.

It's still my favorite DS game. I can't recommend it more. That and Ossu, Tatakae, Ouendan (don't let the import thing scare you off, it's fun for normal people). There's an english version called Elite Beat Agents but it's not quite as fun.

I fully recommend the following
Clubhouse Games - Just for the fact that it has 40 or so different games on it, full on 7 player multiplayer, and its just a good pick me up game for when you arent playing something else. I actually got my partner hooked on it and he has never touched a DS til now. I gave him my Phat when I picked up my DSi, and that stays in it at all times now.
Final Fantasy III - I absolutely love this version, its just the perfect amount of grindin' and the new visuals on it make it very easy on the eye.
Animal Crossing Wild World - Although there arent too many people playing it online anymore, you can try to get a resurrection thread going if you want to play this online, other than that it has enough open ended stuff to keep you busy for a while, and every day something new pops up.
Pokemon Platinum - In my opinion the best out of the three Poke' Games on the DS. If you havent played a Pokemon game start off with this one and you should be good to go.
Magicians Quest : Mysterious Times - This is basically Animal Crossing with Wizards, I am currently playing thru this right now, and I am actually enjoying it alot. I am a HUGE Animal Crossing fanatic so when I saw this I knew I had to get it. This game has a bit more to offer than AC:WW, but AC will always be better no matter what.
The World Ends With You - An awesome RPG, with some awesome play mechanics, if you can find it, grab it. At first the game pissed me off to no end, then it "clicked" with me and I could not put it down for a long time.

All of the above games I have owned and personally recommend. Not all of these games are AA titles though, so use your best judgement. And games like Contact and Lost Magic are not universally appreciated.

I will echo the recommendations of Kirby Canvas Curse. Not only is it an amazingly fun game, but its something that couldn't be done at all (or at least, not nearly as well) on any other system. Its sad that I have yet to see a game that justifies the DS's touch screen as well as this game.
The guy listing GBA games for DS/DSlite owners forgot the Mega Man Zero. My recollection is fuzzy, but I recall 2 being the best of the bunch. I think any one of them would be better than either of the (blah) ZX games.
Unless you are already a big SOTN-style Castlevania fan, I would just get Order Of Ecclesia, as everyone seems to think its the best of the bunch. The games are pretty samey, and if you play them all, you may be burned out on the gameplay by the time you get to OOE.

I highly recommend Dementium: The Ward and Moon. Dementium is a cross between 1st person shooter and survival horror and Moon is a very nice handheld 1st person shooter.
Ninja Gaiden Dragon Sword is probably the best DS game I've played so far. It's fun and it offers some nice challenge.
Summon Night Twin Age is also a good title to pick up. It doesn't get talked about enough and I was addicted to it when I first played it.
Dragon Quest Heroes: Rocket Slime is a lovely little action RPG that deserves more attention.
Castlevania: Order of Ecclesia is the best platformer on the system.
The first three Phoenix Wright games are also amazing.

Now I didn't get very far into this, perhaps only 3 hours or so but I have a very short attention span so errr. Ok, right, the game was very charming - in a quirky humorous way that [strike]you[/strike] I don't see too often. Sort of like Zelda meets Zeno Clash(?)
I feel bad that a lot of games are sitting on my shelf, but a lot more heart ache goes out to Freshly Picked Tingle then to say metal gear acid or ninja town (which was actually pretty fun..shit)
No list is complete nor library of quality without GTA: Chinatown Wars.
This is simply one of the best and most technically astounding games for the DS.
Take a classic style GTA game, blend in the good story-telling of the recent games, mix in some elegant touch-screen controls and minigames (like car jacking! 5 seconds of heart pounding fun!) and add a dash of DrugWars as you buy and sell drugs around town for extra cash.
This game is a superb blend. I played it during my recent trip constantly (thats saying a lot since I had my brand new iPhone with me too to play with).
Satanic Jesus, you are missing one of the best DS games on that list.
Also, get a DSi if you don't have one. There is a lot to love in it. You wouldn't think a camera, some nifty sound software, and an online store add much, but it does!

I used to love the Yu-Gi-Oh! TCG so much when I was younger, but then I just got fed up with some of the bullshit cards/combos they had. I just can't bring myself to play even a video game version of it anymore.

I recommend getting at least one good Rogue-Like for the system. Shiren The Wanderer is the local favorite on the PA boards, but I haven't acquired it yet. I do know that when these boards declare en masse that something is quality, I don't think I've ever disagreed. By all accounts it is brutally hard, which is par for the course on a Rogue-Like.
If you'd like to experience a more forgiving, quality Rogue-Like I recommend the Izuna series. Difficult, but not as punishing as Shiren, with light humor (mainly about boobs, which is a big positive IMO), and a story that DOESN'T have anything to do with saving the world (another big plus IMO). The first Izuna all takes place in one village, has a great little system for buffing and grinding up weapons and scales up the difficulty very well. The last dungeon is the true meat of the game. The second Izuna adds an Overworld map, more dungeons, more quests, a bigger story, about a dozen new playable(!) characters and a tag team system complete with tag team attacks. Still hard, but like the previous game light hearted and fun times. Playing the first game isn't necessary to really enjoy the second one either, which is good, because it's hard to get a copy of the original.

Instead of wishing for a sequel, get Soul Bubbles. I've only played the demo of it while it was on the Wii's Nintendo Channel, but I totally loved it.
It's not an easy game to find, though. It was a Toys R Us exclusive when it came out quite a while ago.

Meteos is probably my absolute favorite DS game for local multiplayer. It was incredible.
It's also got a good amount of single-player content, but I think there's a bunch of better DS games in that regard, now.

If you'd like to experience a more forgiving, quality Rogue-Like I recommend the Izuna series. Difficult, but not as punishing as Shiren, with light humor (mainly about boobs, which is a big positive IMO), and a story that DOESN'T have anything to do with saving the world (another big plus IMO). The first Izuna all takes place in one village, has a great little system for buffing and grinding up weapons and scales up the difficulty very well. The last dungeon is the true meat of the game. The second Izuna adds an Overworld map, more dungeons, more quests, a bigger story, about a dozen new playable(!) characters and a tag team system complete with tag team attacks. Still hard, but like the previous game light hearted and fun times. Playing the first game isn't necessary to really enjoy the second one either, which is good, because it's hard to get a copy of the original.
