The Rock Band 3 announcement could - and I stress COULD - be the nail in the coffin.
Rock Band vs. GH3 = Pretty even split amongst most fans of the music genre. After GH3 had been out for awhile, word got around it was balls hard and not fun towards the end, and people took note that RB was doing weekly DLC releases. All in all though, fans were split.
Rock Band 2 vs. THE WORLDS FIRST BAND GAME = Scales slightly tipped in Rock Band's favor. Everything was backwards compatible with previous songs, and GH's touchstrip sucked. The charts for GH were pretty much universally declared inferior, and even bands like Tool that people went nuts over didn't really cause that big a stir once the game actually came out.
Then Rock Band decided to release a full on Beatles game, and a family style expansion pack - and GH pumped out a shitton of randomness on every platform possible - including a game consisting entirely of songs that were found in the series already (albeit not with full band play). They also totally squandered the "exclusive" artists they had. They also are yet to allow full exports from the previous games.
Harmonies + Keyboard/Keytar + any other surprises vs. TURN INTO AN ANIMALLLLLLLLLLL
I know the music game genre is dying as a whole, but this could be it in the battle of the music games.

The big wild-card for this year is how Green Day Rock Band affects things. Will audiences buy the crap out of it? Will it drain energy from the Rock Band franchises as a whole, just like the 47 different Guitar Heroes did for that one? Or will audiences largely ignore it?
Beyond that, I'd say Rock Band 3's actually poised to do well this year, considering its changes are potentially intriguing rather than embarrassing. Also Pig Hero is flushing the "oh hey let's play this at a party" thing down the toilet, which was a huge chunk of its appeal.
