No, HTML5 still needs Flash or Silverlight or something to avoid the hell that is developing a non-trivial app in Javascript. I do not give a flying fuck how fancy the HTML5 DOM is or that it has an integrated video player or whatnot. If I have to use Javascript to create any substantial functionality it is still an utter failure.
jQuery is that something. There are other frameworks as well. There is absolutely no need to jump all the way to proprietary binary browser plugins.
JQuery, and yes I have developed with it professionaly, is a hacked up workaround to the fundamental flaws of Javascript. What they have acomplished is remarkable but it still pales in comparison to the benefits of any real programming language. While I don't much like proprietary plugins their benefits in terms of scalability, ease of development, performance, language features make them a regretable but vital workaround to the aweful and fundamental platform flaws inherent to in browser Javascript. ok, so Actionscript 3 isn't great but still at least it is typed. And Silverlight uses full on C# 3.5.
and that is assuming that with HTML5 they actually improve the aweful state of cross-browser bullshit that takes up the majority of web-dev work nowadays. There is no way they are going to get Chrome, Firefox, Safari, IE etc... to play nice with each other in terms of CSS rules.
I'll give you ease of development and testing, but performance, at least of Flash, is downright AWFUL. And if you have the gall to use a Mac? Not only will your website take up 80% CPU, it'll crash the entire browser frequently unless the user is on a 64 bit system (in which case just your page goes dead).
And if you want your site to work on mobile platforms? You don't have much of an advantage there. Of the mobile platforms that even support flash, you're not going to get anywhere the level of performance or feature set of the full PC plugin. Silverlight support is even more rare.
At this point, the drawbacks are really adding up.
In terms of browser compatibility on CSS, Chrome and Safari share a rendering engine, so they're good. Opera has never been a problem for me. Firefox has a few quirks but nothing major. Internet Explorer is always the troublemaker, and with IE8, even it is starting to change, bit by bit.
I think you'll see Javascript/CSS problems clear up over the next 2 years, browser support will finally fall in line, frameworks will improve, and plugins will go the way of the dinosaur. I'll eat my own cock, etc, etc.

It's my favorite series outside of House at the moment, and once I have enough money I am buying that car and putting an SS engine in it from a similar model which will fit the engine bay.
