Having put the last episode of Ashes to Ashes to bed (Oh My God how awesome was that?) I'm now left with a hole that needs filling. My wife and I need new dinner time DVD viewing, can you help with some recommendations?

House is another show which is truly great, it is very strongly character driven and has more in common with a detective show than a medical drama, though it is really neither and is probably one of the best TV shows I have ever seen. Very funny at times, at other times it is extremely moving and has suprising depth.
Chuck I have just started watching recently and it is great. It's funny and fun and really worth the effort. It doesn't really fit your themes other than great character stuff and humour. Everyone should check out this show because it is so cool.
be advised that it got canceled with no real closure

An anecdote: my extended family gets together for like a week each Christmas, and one time I brought along Freaks and Geeks because my sister had never seen it. By episode four we were forced to start over because the entire family (like, all 20+ of them) was now watching it with us and loving it and people were demanding to see the episodes they missed. We all burned through the entire series in a few days.
Whites just started, and it's pretty fantastic. I like Sherlock, too, though the 2nd episode is terrible and it's a touch hokey sometimes, but it's a great lighthearted show.
House is great, especially the earlier seasons. It's basically Sherlock Holmes with more drug abuse and some really great ethereal aspects enter the show at one point. Season 6 is trash, however. The seventh starts poorly but is shaping up nicely.
Dexter is another good one. I'm catching up with it, and while slow at times, John lithgow plays a main role in season 4. It's going to be worth watching just for that.
Buffy and Angel meet every one of your criteria perfectly and you have like 12 seasons so that's a good start

Oh yes, Dexter is excellent. Season 4 is just fantastic television. I'd say it counts for Strong Character Driven, Dark Humor, and Things That Are Awesome.
The rest of Dexter is equally awesome.
Also, i know I already mentioned it, but I really feel the need to push Pushing Daisies. I watched it on Netflix instant watch on a whim and it was just so amazing. I wish I had seen it when it was on tv, but it also fits all of your criteria and then some.
Oh man yes, Farscape is great. Everyone should watch this show, and enjoy just how trippy and hilarious it can be while simultaneously being an awesome space adventure where every alien speaks with an Australian accent. Plus Ben Browder and Claudia Black so hey, something for everyone.

I think some of the playful banter (read: sexual tension) between Scully and Mulder would qualify as humor.

It's dated absurdly well, and things like Twin Peaks probably wouldn't exist without it

Only 2 brilliant seasons were made. One of the few shows like this i've rewatched and still really enjoyed.
Also, the prices on Star Trek seasons have done down, so that may work for you.

The new Dr Who series is fantastic. You mentioned firefly so I figured supernatural was not a hard requirement.
Life on Mars had an american version as well which is pretty good. I actually have yet to watch the British version. Harvey Keitel plays Gene Hunt.
The Dresden Files is a shortly lived series based on the books which hits a few items on your list.
