Get some sleep, check your mirrors often, and don't drive aggressively, like accelerating to get into a different lane when there's a car next to you (you should decelerate and go in behind the car instead). Pray like hell that you know how to parallel park if you're going to be required to do that.
But the biggest thing? Calm down, because it really isn't that bad, honest.
unless the examiner seems like a total ass... chat to him as much as possible before the test. Try to make him a human being rather than an examiner. I had my test a few weeks back and there was like a 2 minute walk from the test center to the car, and i talked the whole way there, we chatted about why i was learning to drive, a stupid new road they're building, life in general, the weather etc... just to get a feel for the guy and what he's about. by the time we sat down in the car i already felt reasonably comfortable with him, and had a measure of his personality somewhat. It made me much happier anyway, just thought i'd suggest it! It also gives them a chance to measure what kind of person they think you are, so if you chat calmy and politely, then drive calmly and politely... it backs it up in their heads.
also... it's better to be marked down for hesitation / slowness than for hitting someone else... hesitation is just points scored down.. if you cause someone else to swerve or whatever... you've just failed. So if you're not sure if you'll fit in that gap etc... wait till you're sure.
also... don't be afraid to pull out and have another go when parking etc...
mirrors, mirrors, mirrors, everything you do, no matter how small, check them, i got told off all the time for forgetting to check my mirrors before indicating.

dont be worried or surprised if you can't tell how well you're doing until the end of the test

mine was pretty quiet until the end when i found out all I got were 5 out of 75 demerits (for "signaling too early" - which I couldn't even remember doing)

It's as much of a stress test as it as it is a test of your ability. Treat it like it's no different then having your mum or dad telling you where to go/what to do when you were learning. It's really all about how you handle yourself, so don't get all freaked out.
You'll be fine. Just remember if your parents taught you to drive they may have shown you some tricks that are technically against the rules. (Mine never bothered to tell me that turning left into the right-hand land is a no-no, for example)
Remember what you learned in Drivers' Ed. Pay attention to the rules of the road. Stay within the speed limit. Don't blow through stop lights. Check when you're turning that you're turning the right way if it's a one way road. Look for "No right turn on red/no turn" signs at stoplights. Things like that. Just drive responsibly and defensively. It only takes a little while and then you'll be done and have your license.
Remember that so long as you don't do any of the "automatic fail," things then you would have to actually try to do enough of the little things to really fail a drivers' test.
Thanks a lot for all the suggestions. Im going to bed early tonight and taking some asprin before the test to cool the nerves. I'm a pretty good driver on the road, but since this is on a closed course with only other test takers on the course. I've memorized all the basic functions like lights, whipers, fluids, etc. The only real thing im worried about is parallel parking. The whole, if you hit the cone you fail is unsettling.
What main things should I focus on during these parts?
Check your mirrors constantly. Probably even more so than you regularly would. I remembered I failed my first time because I didn't check enough, even though I checked just as much as I normally do.
if you have any time to do some practice... heres something my instructor taught me which is ridiculously good and has never failed me once... it won't help if you're doing it on cones thoguh :S
drive past the car , about a car door length away from it until you yourself are level with the front bumper.(ie you look out of your side window and are looking across the very front of the car)
shift to reverse and slowly go back, looking over your shoulder out of your passenger side rear window,
when you see the back of the other car appear in this window you do one full turn towards the kerb (meaning a 360 rotation of the wheel, not full lock). you hold the steering there and let the car roll backwards slowly,
when the front of your car is level with the back of theirs you do 2 full turns away from the kerb, that should bring you in nicely behind them,
once you feel you're level you can do one full turn back towards the kerb to level up.
If you turn too quickly or slowly you might need to just wind a little bit of extra lock on or off, but as a general guide it works really well
i was terrified about parallel parking, but once he taught me that... i was so much happier about it, knowing there's a formula that really works helped me a lot.

Exactly. I checked all three mirrors trying to be obvious as possible about it. At the end of the test, the instructor told me i lost points for not checking my mirrors before i merged into traffic. Then again they also failed me for speeding when i was doing 27 i think in a 25 zone (didn't see a sign so i assumed it was 25) when the speed limit was actually 35. Also, don't forget to use your turn singles when parallel parking. I used mine going in but when the instructor said "good, now lets move on" i was thinking that the parking portion of the test was over and i wasnt being graded on it anymore so i lost points for not using my signal on the way out.

What was suggested to me when I did my driving test was to actually wear a ballcap and exaggerate your mirror checking so it's painfully obvious you're doing it.
My only specific advice. Otherwise, as a slew of others have said, relax, be courteous to the instructor, and just drive.
I just thought of something else. Turn off your radio and cell phone before the instructor even gets in the car. Personally, i cant stand driving without background noise but you dont want any distractions while your taking the test.
From experience, I failed my first test because I didn't look over my shoulder when merging into a bike lane to make a right turn. The only other mistake I made was not signaling after parking alongside the road. Listen to what they said and don't make the same mistakes I did.

Chances are you already know this but only use one foot to drive. When i first started out learning to drive i was nervous as hell and wanted to keep a foot on the brake at all times. No one ever told me this was wrong and it turned into one hell of a tough habit to break.
Yeah I almost failed mine for two reasons:
1) Was driving my mom's Expedition.
2) Was paranoid as hell. The person who was giving me the test kept telling me to calm down and laughed at how nervous I was.
Also if possible, drive a small car. Makes things so much easier.
And be blatantly obvious about it. Some instructors will assume you didn't check unless your head moves.
When you check your mirrors, actually say quietly to youself "mirrors". This will make it obvious.
I failed my first test because I fucked up one section (here there are 7 sections and you need to get 6 out of 7 sections to pass) and freaked the fuck out. This caused me to stall the engine fuck up my parking and I failed because I let that one fuck up affect me for the rest of the trip.
Don't do that. If you make a mistake just let it go, it's already done and you can't go back and fix it you just gotta deal.
If this is the DMV test, there is a written portion you should worry more about, because it's completely pedantic. E.g. how many alcoholic drinks can a 180 pound person consume before their Blood Alcohol Content is over the legal limit, etc. You may also be surprised that they will actually ask how many feet from the intersection you are allowed to park, even though 99.99% of intersections where it would matter are marked with signs and/or curb paint. They still ask this stupid crap.

I think that may just depend on where you live. The only test I had to take was a multiple choice test on a touch screen computer for my permit. Half the questons were stupid stuff like should you drink then drive?, should you smoke weed and drive? ect.
