A question so easy that lock is not too far away...
I'm debating purchasing Rock Band or Guitar Hero for the Wii, problem is I'm not too sure how into it my wife and I would be, so renting would be the obvious choice... Does Blockbuster rent out the Guitar and such when they rent the game too, or no?
You should call them before you go to see if they have it in stock. I would imagine that, since you need the guitar to play the game, they would rent both -- if they rent the game at all.
If your wife is the prototypical casual gamer, she'll more than likely enjoy Rock Band more. Actually, I can't think of any reason she would like GH over RB. Maybe if she prefers the songs from GH, but that seems unlikely.
As far as whether or not they rent out guitars, I think it would be best to call your closest rental store.
Last time I was at Hollywood Video, they rented Guitar Hero and the guitar controller separately, so if you wanted both it was the cost of two game rentals. This may have changed; I haven't been for months.
I have no idea on the rental situation. I second going into a store to try them out. If that's still not an option, such as because the instruments in the store are beat to hell and barely work, which is all too common, then I say Rock Band. Rock Band has a wider variety of ways to play, so perhaps she wouldn't like the guitar bit, but would still like the singing or the drums. The guitar parts in Rock Band are mostly easier than the Guitar Hero games, so I find more people who enjoy it more because they can play it more easily. Rock Band also has more accessible music that is enjoyed by more people. I have heard a lot of people complain about the track lists for the Guitar Hero games, saying that there's too much hard rock and heavy metal. The base Rock Band track list has more pop/radio rock type songs and the metal that is in it is not as extreme as some of the Guitar Hero metal tracks. While I have not checked up on the Guitar Hero 3 dlc, Rock Band has more and better DLC than Guitar Hero 2 got.

I've never seen a Blockbuster rent out guitars and such, but I've only been to about three in the past 3 years. I think your best bet is to try and go to a retailer like someone suggested and play them there or go to a friends who might have them. I've never seen Rock Band out for demo but I'm sure there are some somewhere, and there definitely will be when Rock Band 2/Guitar Hero:WT hits.
By the way, the next Guitar Hero is following the Rock Band model (full band), just so you are aware.

OR, and i am less fond of this choice, go to a large retailer. Most of them should have displays set up. If not for Rock Band, then for Guitar Hero.
