Advice: Computer Virus on Home Network

My wife's computer got a virus. Malwarebytes got rid the obvious visible problems, but there's still something definitely wrong, as MSE got disabled/uninstalled, and for some who knows what the hell reason, some weird music started getting piped to my wife's computer.

Fine, nuke it from orbit. It's a PITA, but it's the right thing to do.

My question, for you fine computer friends, is how much do I need to worry about the other computers on the home network.
I run software firewalls on all computers.
My router is a fairly new router that I keep upgraded (Netgear...probably should ddwrt it someday).
MSE/Malwarebytes show nothing on the other computers.

I think I'm safe, but I'd like to make sure I'm doing due diligence.

    Edit: And a scan with MSE just revealed something called sirefef sitting around. Ooo fun, yes, after my wife moves all her docs, the computer gets nuked.

    I don't think any other computers on the network I need to worry about though.
