I got a job there and would like to know all the tricks and tips on best ways of getting there.
What routes have most traffic, how much are all the tolls, or should I take public transport and how much is that, etc.

You might be best off with public transportation. Take the subway to Penn Station, and from there a train straight to EWR. Once there, just take the AirTrain monorail and you can get to whatever terminal you need to. Just keep in mind that the train from Penn + the AirTrain is about $15 each way. The train from Penn into Newark is about half an hour too, so add that to your commute from Queens.
Public transportation is probably the least-hassle way of getting there, and like RFX says, there's a direct train from NY Penn to Newark Liberty (one stop past Newark Penn--takes about 25 min), and then the AirTrain monorail from the Newark Liberty train station to Newark Liberty airport (takes about 5 min).
If you really want to drive though, it's heavily dependent on what time you go. I would say to avoid at all costs the BQE and the Staten Island Expressway (I-278). Those roads have bad traffic pretty much all day everyday, and are under construction 24/7 (and will be forever). That means, don't take the Verrazzano bridge through Staten island to get there, even if it "seems" like the best route. The best way is to go through Manhattan and take the Holland Tunnel, then when you get into NJ take Rt. 1-9 South (express) all the way to the airport (Total tolls paid: $0, assuming you took a bridge to get into Manhattan). Taking the NJ Turnpike from the Holland Tunnel might also be faster, but there's a toll involved with that. This is also heavily dependent on traffic conditions, and the Lincoln tunnel might be a little faster depending on what time of day you go--check maps.google.com for realtime traffic right before you leave to confirm. But in my experience, the Holland is usually the fastest way out of the city.
