Lately I've been doing a lot of texturing for my mod, thus I'm at the PC uninterrupted for huge blocks of time (4-6 hours at least). I used to just listen to music, and built up a huge library...but I'd like to be a bit more constructive and I started listening to podcasts, gaming related ones (ideally ones focusing more on game design/business). A quick list of the ones I've blown through;
-Penny Arcade
-Unknown Worlds (eg. the Natural Selection guys)
-Gamasutra's GDC podcasts (all the free ones, plus the paid ones I thought would be of interest)
-University of California, foundations of game design (a professor podcasted most of his entire class)
-Microsoft's podcasts from their developer conference
-Game Theory
And right now I'm trying some of the PC Gamer ones again (but previously found it to lack substance...).
Any suggestions for others?
Hell, if you just know a really cool podcast not like the above go ahead and suggest...I'm really tapped out. :P

I used to listen to the Snack-Bar Games podcast, but they went from one show a week to "whenever they feel like it," so it's not really worth having on the roll anymore.
Well, technically, the aforementioned GFW Radio is a 1up podcast. And I second it and any other 1up podcast around. They're all pretty great, although none of them really focus on the design side of gaming per se.
In terms of just plain good podcasts you might want to look into This American Life.
