Yes, play the game, it's how I got into it. Then for further curiosity check the comprehensive Wiki article and the official website background articles. But it's a lot better if you play the game and see stuff in action, and then read about them in more detail.

Yeah, I knew a bit about the Space Marines from the table top game, but after playing DoW, you really get a feel for the lore and the whole universe. Especially from the first few missions in the Original Campaign. From there, just read up a few wiki articles and your golden on how the Imperium of Man works.

Those guardsmen need to stop damaging morale what with their cowardly retreating. If a few have to die to a commissar's pistol to better encourage the rest to fight harder in the name of the emperor, then so be it.

Realized today the only races I haven't beat the DOW:DC campaign with are the Orks and IG. Ugh.
